SmartGlow™ Pre Stain, for Nucleic Acid Electrophoresis Gels, 20,000X

SmartGlow™ Pre Stain can be used the same way as Ethidium Bromide for agarose and polyacrylamide gel electrophoresis. The stain emits green fluorescence when bound to dsDNA or ssDNA and emits red fluorescence when bound to RNA.

SmartGlow™ stain has two excitation peaks at approximately 290nm (UV light) and 490nm (Blue light).

Protocol:

1. Prepare 100ml of agarose or polyacrylamide solution

2. Add 5ul of SmartGlow™ stain to the gel solution.

3. Mix gently to and make sure there are no air bubbles in the mixture

4. Allow the solution to cool to about 60°C to 70°C then pour the gel(s).

5. For enhanced results, you can optionally add 5ul of SmartGlow™ stain per 100ml of the running buffer. Adding the stain to the running buffer can help in increasing sensitivity and detecting small quantities of nucleic acid.

6. After the separation is complete, view the gel using a UV or blue light illuminator.

What is the shipment/storage temperature?:

For the maximum performance and stability SmartGlow should be stored at +4°C and in the dark. This item must ship cold and follows our cold ship policy which you can read here. 

What is the shelf life of SmartGlow DNA Stain?:

Minimum 2 years when stored properly at 4°C.

Can SmartGlow Stain be diluted?:

SmartGlow solution can be diluted in the ultrapure water.

Can SmartGlow be used for RNA as well as DNA?:

Yes, it can be used for RNA, dsDNA and ssDNA.

Is SmartGlow suitable for UV and blue light illumination?:

Yes, SmartGlow stain is suitable for both UV and blue light illumination.

Can SmartGlow be used with TAE and TBE buffers?:

Yes, when running agarose gel, the electrophoresis buffer may be TAE or TBE buffer.

Is it possible to stain DNA with SmartGlow Stain in agarose gels using the same method as with EtBr (ethidium bromide)?:

Yes, it can be used with the DNA in the gel like EtBr.

SmartGlow has two secondary fluorescence excitation peaks: (~270 nm; ~290 nm) and one strong excitation peak around 490 nm. The fluorescence emission is centered at ~530 nm.

Which filters (wavelength) can be used for SmartGlow imaging?: The filters designed for green dyes (like GFP/SYBR green filter) are the most sensitive. However, EtBr filters and even UV-light without any filter can be used.

Does SmartGlow work with low concentration of DNA?: Yes, it works well with low concentrations of DNA. However, smaller fragments, less than 300 bp may not be as bright as the larger ones (post-staining).

Does SmartGlow interfere with downstream applications?: Avoid leaving SmartView Stain in downstream solution. You may purify the DNA solution with a purification kit.

Can SmartGlow be used for pulse field gel electrophoresis?:

It is not recommended to use SmartGlow for pulse field gel electrophoresis.

Can SmartGlow be used in Southern blots?:

Yes, SmartGlow is compatible with Southern blotting experiments.

Does the stain interfere with recovery of DNA from the stained gel once it is run?:

No, the stain should not cause any problems for further experiments with that DNA.

Does SmartGlow Stain penetrate latex gloves?:

SmartGlow does not penetrate latex gloves.

How can SmartGlow be disposed after use?:

SmartGlow does not create any toxic waste. Therefore, it can be disposed according to laboratory regulations.
